Effect of luteal phase porcine follicular fluid (pFF) from medium size (4-6 mm diameter) on primary porcine oviductal epithelial cell (POEC) viability was evaluated by MTT assay and analyzed by one-way ANOVA. POEC were cultured in M199 supplement with 10% fetal calf serum (FCS) in high humidity with 5% CO2 at 37°C for 96 h (n=6). After culture time, POEC was spindle shape and expansion. Then, they were treated with pFF from medium size follicle at concentration of 2 μg, 4 μg, 20 μg, 40 μg, 200 μg, 400 μg, and 500 μg, for 24 h. We found percentage of cell viability at concentration of 2 μg, 4 μg, 20 μg, 40 μg, 200 μg, and 400 μg showed slight increase in cell viability. Meanwhile, percentage of cell viability at 500 μg was exhibit higher than positive control (medium with 10% FCS+ hormone) and control group but not significant (p>0.05). Even though this finding is basic research, but the results would be important for use pFF as replacement of FCS and hormone in culture medium, in the future study.
